The three men in the vehicle suffered serious injuries and were taken to Crosshouse Hospital.
The 32-year-old driver of the car died a short time later at the hospital, officers confirmed on Sunday.
The other two men, aged 24 and 25, remain in the care of medics at Crosshouse.
